Actually, i am trying to develop the related substances method for paracetamol oral suspension in RP HPLC and i am getting small peak in the diluent at the RT time of paracetamol. I run the blank with empty vial and no peak appeared. My Mobile phase is 4.2g of Na2HPO4 and 2 g of NaH2PO4 and methanol as mobile phase B. and flow rate is 1.5ml/min and running the standard solution i am getting peak in diluent. I washed the column from 95%H2O and 5 Methanol and then 50 50 H2O and Methanol and later 95% Methanol for 15 min and when run the injection peak disappeared and again appeared can anyone please help me how to clean the column to get rid of the peak in diluent.
It is probably a gradient peak (difference in UV and RI) due to the switch over from 100% phosphate buffer to 100% Methanol. Try using 5-10% of the other Mobile phase in the original Mobile phase. Ensure the diluent sample is diluted 1/10 or 1/5 in Mobile phase A just prior to injection Increase the injection volume to compensate).

There are a 'lot' of chromatographic methods on the internet. Just select one and dilute your sample until it is on the linearity curve.
