After acid hydrolysis and incubation in the dark using Taka-diastase enzyme, 30% sulphuric acid was added to stop the enzymatic reaction. The solution was diluted to the mark and some of the solution was filtered and diluted with methanol and water before injected into HPLC vial.
I have tried to search for answers online but this come to me: methanol is used to precipitate protein. May I ask if this true?

Methanol or ethanol can be used to precipitate proteins, indeed. Just as high salt solutions, read WIKI https://en.wikipedia.org/wiki/Protein_precipitation#:~:text=Salting%20out%20is%20the%20most,and%20increases%20protein%E2%80%93protein%20interactions.
What method would be best depends on the proteins in your sample, the sample matrix and last but not least the HPLC mobile phase you intend to use.
Ultrafiltration or gelchromatography might be usefull as well.

Tan... From the physical point of view methanol is miscible with water. Thus, its addition to a clear solution will replace the solute and interacts with water. The equilibrium of the component with water will thus be broken and precipitate. Regards
Methonol , chloroform and water is used to precipitate the protein, which contains lipids, nucleic acids.
