Measuring mitochondrial activity and ROS from in vivo samples?

I am looking to isolate samples of mitochondria from an in vivo experiment, but would like to know the best way to do so. Most of the publications I have found recommend isolating the mitochondria for measuring activity, or performing IHC targeting 4-HNE to examine ROS production. Any recommendations as far as more specifics or other directions to use?

I propose to measure directly some ROS biomarkers after isolating mitochondria. But ROS can differe depending on the nature of isolated cells. What is the cell you isolated?
