Refined wheat flour was analyzed for total dietary fiber, that is, insoluble and soluble fractions, by enzymatic method using the Megazyme Kit. According to the results:
Total D.F. = 0.5 %
Soluble D. F. = 1.3 %
Therefore, Insoluble D. F. = - 0.8 %
Is it possible that insoluble fraction be negative? or is there a mistake during the assay?

i am the frequent user for various enzymatic and flavoring constituents for food concern. TDF is the collective result of soluble and insoluble dietary fibers.
But in your case, you have done with some calculation mistake, may be in minutes or some typographical error is there
Thank you all for your answers. Indeed, there is a mistake. Some papers showed solely TDF and SDF, instead or together with IDF.
From the example, unless the SDF doesn't decrease, it will indicate a procedure error because of the sum of both SDF and IDF.

One additional question
For total dietary fiber (TDF):
If two food samples of the same variety, for example, Manitoba wheat, are submitted to the enzymatic-gravimetric method for DF, and the results are different:
1. Manitoba wheat TDF: 2.5 %
2. Manitoba wheat TDF: 3.9 %
What would be the explanation? Considering that both varieties were analyzed with all their physical components, and harvested in the same growing conditions.
