I was wondering if anyone out there works with macrophages and how they prepare them for assays. Specifically, I am interested in knowing if there is an intrinsic difference between non-adherent macrophages and those that become attached to the bottom of the culture flask and why one would use one or the other for assays. Also wondering if anyone has used ultra-low attachment dishes to prevent macrophages from settling down and becoming attached? Would this homogenize the population to any extent?

In our protocols after thaw, we let the cells sit in a 6 well plate for 3 days prior to use and only collect what's floating. We tend to lose a lot of cells doing this. So I guess my question is what is the difference between attached and non-adherent macrophages since they were already differentiated earlier?
