Does anyone know of an lysate buffer without detergent (like Triton-X or IGEPAL CA630) that can be used in order to lyse human HEK293 cells?
The reason why im asking is that the detergent in the buffer is disrupting my assay later on. Dialysis did not work.

You might want to try Celllytic M from Sigma. I am not sure if it has any detergent as Sigma doesn't provide the recipe but you can email them. This lysis buffer is compatible with a wide range of downstream assays.

Perhaps Millipore's ProteoExtract transmembrane protein extraction kit? You might also try hypotonic lysis and/or sonicate cells.
Awesome guys, I will look into the buffers you recommended.
I have also heard that freeze thawing the cells with dry ice and ethanol could be a suitable method.
