I would like to know why LX2 cells produce a large number of lipid droplets during passages and they show a activated hepatic stellate cell morphology.

Here is the procedure I used to culture LX2 cells.

1. I released the LX2 cell stock in DMEM containing 10% FBS and Penicillin-Streptomycin medium.

2. Next day I removed the media and added DMEM containing 2% FBS and Penicillin-Streptomycin medium.

3. After cells are about 70-80% confluent, I sub culture them in DMEM containing 2% FBS and Penicillin-Streptomycin medium.

After, 2-3 passages LX2 cells show a higher number of lipid droplets and they have a morphology similar to activated HSCs.
