We found that our strains of Fusarium graminearum progressively lost their ability to produce mycotoxins (DON- ADON) on synthetic media such as PDA or Czapek-yeast extract. After a few months using the strains, no mycotoxins were produced anymore, whereas they produced some at the beginning. Any clue on how we could recover the mycotoxin production ability of our strains (specific media, treatment...)?
Hi, Sub-culturing for Fusarium spp cause a mutation or loss the pathogenicity of the fungus, and this is a problem. One of the solutions is grow the fungus in same substrate that you isolate the fungus from. Or go to grow the fungus from the original vial that you store the fungus if you have.
it is true , subcultures in vitro make Fusarium lose its ability to synthesize mycotoxins. As said sir oadi, you have to go back to the original strain that was stored
There may be inhibitory action of the substrate at a given concentration to the growth of fusarium species. I am not sure but can you regularly remove DON. I know it might not be possible. If possible, you may sustain with the process.
Infect the host plant(s) with strain(s) and make a re-isolation. Sometimes it helps.
