my current project is somehow related to ISR (Integrated stress response) stress pathways, I am looking for some decent antibodies that detects phospho-HRI (heme-regulated inhibitor kinase). I have found some but only detect total forms of HRI. Any suggestions?
Do you know which phosphorylated amino acids at which position you need to search for? That is essential. With the anti total HRI, you then may quantify the total amount of the protein and normalize your results, so you need both antibodies, one specific for the signal you're after, and one for the total protein.
In case of pTyr, you'll be fine with the PY20 monoclonal (it detects virtually all phosphotyrosines in all proteins), if the phosphorylation you're looking for is pSer or pThr, you'll need one or more specific antibodies against the whole motifs (a few adjacent amino acids left and right of the phosphorylation sites). If you expect to need large amounts, consider to have one custom-made.
