I am studying T-Cell activation and while we have markers for T-Cell activation that work reliably well we are looking for the population of T-Cells that were not activated despite being exposed to CD3/CD28 beads. Currently we are using an HLADR marker along with CD39 to look for a double negative. I am wondering if anyone else has attempted to do this that might have more input or some references to look at.
Hi Raymond, T cell activation is really well described in many articles and the timing of different activation markers are also defined.
In general you can assess the expression of CD69, the expression of which increases in the first 24 hours after activation. At 48 hours you can analyze the expression level of CD25, the IL-2 alpha receptor, which is upregulated following activation and is used for the uptake of IL-2 itself that is produced and released by the activated T lymphocyte and which, in turn, is used for clonal expansion. The absence of these two markers maybe could be helpful to you
CD 69 is the most common and earliest marker to confirm T cell activation. Besides, CD 25 is routinely use as murine T cell activation marker
