What factors should one consider while selecting concentrations of the internal solutions in the micropipette used in patch clamp?
What Ion concentrations are optimum for mEPSC and mIPSC?

The choice of intracellular solution should be made taking into considerations the overall goal of the experiment. I.e. if you are interested in measuring membrane excitability, you should avoid gluconate salts. For getting better space clamp, you should add salts blocking leak currents (i.e. Cs). For GABA currents, Cl reach internal solution are recommended. Unfortunately, I am no aware of a single article presenting this important topic, with explanations. Typically researchers have their solution cook-books and get this information from papers. I would be happy to answer specific questions concerning to your needs. S -
