I have some amount of experience using lentiviruses for protein expression and knockdown but I'm considering AAV as an alternative. I'm curious to hear from researchers who have used both lentivirus and adeno-associated virus for expression of proteins or RNAi... What are the pros and cons of working with each system? Obviously this will depend on the intended application. I work with primary neuron cultures from hippocampus or cortex, but any insights on terminally differentiated cells would be helpful.
1) is there any difference in the transduction efficiency between the two?
2) is there any difference in the speed, ease, or reproducibility of virion generation methodology?
3) do they differ in the level of protein expression or efficacy of RNAi
If anyone has worked extensively with AAV shRNA expression, do you have a preferred vector plasmid?
