To assess root colonization by AMF, I want to use the grid-line intersect method (Giovannetti & Mosse, 1980) or even the magnified intersection method (McGonigle et al., 1990).
I understand the principle of the methods. But how can I best train my eyes to recognize mycorrhizal from non mycorrhizal structures? I have found articles, handbooks and manuals detailing the methods (and others) but I cannot follow it if I do not know what to count or not as a mycorrhizal structure. I am looking for a person, course, book or online resource that could teach me this skill.
If it is relevant: I am working on wheat roots sampled about one week before harvest.
I would be thankful for any help!

Gallou Adrien I have been in contact with Prof. Declerck from UCL who helped me interpreting a few photographs.
Gonzalo Galileo Rivas Platero I had in fact already found that website, and it is full of resources indeed. Although I mostly needed to train my eye for the less obvious cases, where structures are not always as easily recognized as in the pictures. Since then I realized that some of the difficulty I was confronted with came from a suboptimal clearing.
Mohammed Fayyadh, your file certainly helps me with calibrating my method (especially the clearing part, where I have been somewhat too cautious at first).
And finally Manju M. Gupta thank you for the hint. I had already seen mention of that book on the website that Gonzalo Galileo Rivas Platero recommended. But I had not thought of looking for the full text on ResearchGate, where I am still quite a newbie.

I am surprised to see the lengthwise arrangement of roots, as I had seen mounted slides with the roots segments perpendicular to the slide length. But I appreciate the idea of letting the mountant flow around the roots, as I have been struggling myself with the issue of spreading the mountant through my sample without overflowing it. I will definitely try this out.
I also see that your diagram shows root segments of varying lengths, although I had read that I should use 1 cm long segments. I must say, I am sometimes confronted with shorter root fragments and I would appreciate not having to disregard them, especially when my sample is a little poor.
