I'm working on a mesenchymal stem cell culture isolated from rat bone marrow and I want to yield a specific MSCs count from passage 3 . I want to know the yield of cells in passage 3 to know how many rats I want to isolate MSCs from its bone marrow to get the count I want at final.

1. It will depend on the isolation procedure. Once you isolate BMMSCs, followed by subsequent passaging will give you well adapted and proliferative nature cells in your plate.
2. The doubling time of MSC is in the range of 16-24hrs. Now, if cells gain their best proliferation then you can expect a good amount of cells.
3. However, the culture plate format and seeding number before a particular passage will matter. Since you are focussing on P3 so when it was in P2, what was the cell number, in which plate you are going to seed, and what is doubling time that cells are following?
