Goodmorning everyone!
I am using the IWP-2 inhibitor (IWP-2 inhibits the WNT pathway at the level of the pathway activator Porcupine) on melanoma cell line FO1 with a 20 µM concentration but when I add it to the medium into the wells it is not completely soluble and form visible precipitates, do you have any suggestion to avoid this problem?

Hi Serena,
What are you dissolving your IWP-2 in? For insoluble drugs such as this, I would recommend dissolving in 100% DMSO then adding 20uM to your cell culture in as low a volume as possible. As long as you don't go above 0.5-1% v/v DMSO in the culture media then your cells should be fine. From the datasheet I see IWP-2 is soluble above 5mg/ml in DMSO so this shouldn't be a problem.

Hi Serena, did you subilize the IWP2 in DMSO? you heat during the preocess? what final concentration you prepare your stock? 5 mM is a maximal concentration and the heating is critic.
