All available protocols use the Percoll gradient to isolate T cells, unfortunately we only have Ficoll 1,077 available at the moment. Does anyone experience you with this type of isolation?
Ficoll 1077 is useful for this process. You need tissue preparation before density gradient such as the Percoll assay. And, then you can try Ficoll separation.
After reading discussion and litterature I found that percoll is used to remove T cells as they only wanted Monocytes.
So, if you want T cells from brain tissue, follow the protocol with Ficoll. After you make single cell suspension, you should phenotype the T cells by Flowcytometry before moving towards sorting.
