Currently, I've been working with inmunofluoresce for my thesis, and the aim of my project is to detect the germ cells, in the early stages of embryonic development for an insect. Before doing some essays with embryos, I wanted to try the tecnique with Adults, by marking the stem cells in Testis or ovaries. Am I doing right? could someone recommend a technique for me in order to follow an specific protocol?.

Vasa (DDX4) is also expressed in ESCs. If you are doing this in insects at an embryonic stage past gonad formation, you can certainly use vasa to detect germ cells. Refer to Drosophila papers by Ruth Lehman's lab and Yukiko Yamashita's lab.
