ERM proteins can also be phosphorylated at the conserved T567/T564/558 site by several different kinases other than SLK, e.g. ROCK or PKA, and e.g. Src at non served sites within Ezrin alone. So it depends on what you want to investigate. SLK-mediated ERM phosphorylation/activation or general ERM phosphorylation/activation? Moreover, do you want to inhibit ERM want to interfere at the level of ERM phosphorylation/activation or do you have a specific downstream ERM function in mind that you would like to block?

For the later case there have been some artificial and one natural mutations published within ERM proteins which might do the trick for you. You can either check PubMed for loss-of-function mutants like Ezrin R40L, A129T, R572A or R579A (there is also T567A, but that appears not to behave as expected) or send me a message for some more information.

For the former, there have been two inhibitors published. One specifically against ezrin, which is commercially available, Ezrin Inhibitor, NSC668394 - Calbiochem. However, in our hands this did not work as intended and published, so I would use it carefully and well controlled. Another one specifically against Radixin, quinocarmycin analog DX52-1. I never managed to get my hands on it, though. The group of Georg Kuhn managed to get a sample from the publishing lab (Prof. Gabriel Fenteany), used it and produced promising results. I tried to contact Prof. Fenteany myself but without luck, and I don't know that DX52-1 is commercially available anywhere either.

So in short, no there is currently no readily available, reliable ERM inhibitor available. Your best options at the moment are using knock-down / knock-out / and/or expression of mutant ERM proteins. Just write if you need to discuss more.
