I tried lipofectamine LTX to transfect cerebellar granule neurons, but the efficiency is too low. Actually I used the same method to transfect cortical neurons, and it turned out good efficiency. So what's wrong with this method? Is there any better way to transfect cerebellar granule neurons? I am thinking about the electrotransfection, but it only works in floating cells. Maybe I can try that on the day I am planting the neuron. My only concern here is whether the plasmid can survival for that long, like 15days div?
