Hi dear all, greetings, I want to count the total filamentous fungus from a fermented food sample which contains both yeast and molds. Now I’m using 3M Petriflim for yeast and mold count. My food samples have a high amount of yeast count (6-9 Log CFU/g) and a low amount of filamentous fungus (1-2 Log CFU/g). While I used 3M Petriflim the 10-1 to10-5 dilutions are TNTC (fully occupied by yeast cells only) but a filamentous fungus may be growing at 10-1 to 10-3 dilutions (due to TNTC (Pertiflim fully appeared green in color) the fungus are not able to identify). Anyone could suggest a selective media to grow filamentous fungus only (not with yeast) or any antibiotics or other methods? thank you so much.
@ Srinivasan, you may try with SDA media. The attached file may be useful to you.
You could try adding some fluconazole, most filamentous fungi are not sensitive to this drug.

There is an old but interesting method I still use when I want to isolate filamentous species avoiding yeasts. The method exploits the fact that hyphae can grow trhogh the agar, while yeasts can't. You pour your dilution onto the bottom of a sterilized petri dish (with no medium) and move it arround and let it evaporate (just until there is so little liquid it doesn't moce anymore. Then you pour the agarized medium (at 49 C, just before it get's solid) and cover the bottom. This way, only filamentous fungi will find their way throug it.
Alternatively you can por your dilution on the solidified agar medium and let it dry, and they flip the medium with a spatula (without touching the surface) and wait.
You don't need to add inhibitory substances (they will all affect the outcome) and you'll be eve able to isolate the colonies you want to keep.

I suggest you use crystalline cellulose as a sole carbon source. Hope only your fungus will grow there.
