Roughness is a parameter (or set of parameters) which can be determined by analyzing of a properly acquired line profile.

Yes, an AFM line profile contains information about the RMS roughness of the surface. Keep in mind though, that an AFM image is a convolution of surface and tip shape. Is the size/shape of the tip smaller than the roughness features of the surface then getting that information is pretty straight-forward. If that is not the case, it gets problematic.
