Hello everyone,

We are working on osteoblast cells differentiation from bone marrow cells of the mice, for this we have isolated the bone marrow cells from tibia,femur and adipose tissue. we have been trying to isolate bone marrow cells using a number of methods reported earlier, one of which Maridas, D. E., Rendina-Ruedy, E., Le, P. T., Rosen, C. J. Isolation, Culture, and Differentiation of Bone Marrow Stromal Cells and Osteoclast Progenitors from Mice. J. Vis. Exp. (131), e56750, doi:10.3791/56750 (2018) but did'nt get healthy MSC cells. As we know bone marrow cells are rich in haemopoeitic cells in cmparisons to mesenchymal cells.

our keen interest is Mesenchymal cells because it differentiate into osteoblast cells.

Is there any method from which we can get sufficient number of mesenchymal cell and easy to propagate in a culture.

Culture condition which we have followed:

1) alpha-MEM

2) 1% Non essential amino acids

3) 1% L-Glutamine

4) 0.5% anitibiotics( penicillin and streptomycin)

5) 10% FBS

6) Incubator conditions: 5% CO2 and humidity.
