Actually my CO2 incubator is damaged and I don't know the concentration of CO2 so my MCF7 cells are not reproduced actively. Basically I am using RPMI but it doesn't work good with low CO2. So is there a MEDIA that can work good in this condition?
L-15 media (Sigma L1518) supposedly can be used to grow cells without CO2, but better to get your incubator fixed:-) HEPES is used to prevent rapid shifts in pH when cells are out of a CO2 environment; not sure it would maintain pH in air, but worth a shot-try 25 mM.
Hope you find this beneficial; For culture CO2 environment , you can light a candle inside the incubator then close it , then you will get Co2 environment inside your incubator after the candle goes off , as it will consume all O2 present in your incubator and produce CO2 ,I don't know the concentration of CO2 that will be produced , if enough for your growth media .? butI think it is better to get your incubator fixed.
