I would like to use PBS with Calcium and Magnesium in order to favour keeping my cells adhered. However, the HBSS and PBS with CaCl2 and MgCl2 that I have tried all give me cloudy solutions which makes a mess of FACS.
I think the cloudiness is due to the common ion effect, i.e. producing Ca3(PO4)2 and possibly Mg3(PO4)2, which have relatively low solubilities, so perhaps this is just what happens with this type of buffer?

We use PBD with Ca and Mg from Gibco (just makes life easier) and it is definitively not cloudy.
Hi, Please check the pH of your PBS and HBSS, as it should be less than 7.3. Second I think it's better to autoclave the PBS before adding Calcium and magnesium. Because autoclaving with calcium and magnesium make precipitation.
Thirdly, if you have autoclave PBS with added calcium and magnesium, I will suggest storing at room temperature is better.

The solution is cloudy as prepared using standard protocols, but becomes clear when pH is adjusted to 7.2.
