I would like to use PBS with Calcium and Magnesium in order to favour keeping my cells adhered. However, the HBSS and PBS with CaCl2 and MgCl2 that I have tried all give me cloudy solutions which makes a mess of FACS.
I think the cloudiness is due to the common ion effect, i.e. producing Ca3(PO4)2 and possibly Mg3(PO4)2, which have relatively low solubilities, so perhaps this is just what happens with this type of buffer?
