Hi, I have a Sutter Puller P-97 and I am looking for protocols to pull glass capillaries (final tip diameter > 20 um) for injecting cells into tissues. Could somebody advice on which parameters use (heat, pull, velocity, time) ? thanks
I had to look one time for trouble shooting the same in our lab and came across their manual. There are a basic settings mentioned in the manual to try. http://www.sutter.com/manuals/P-97-INT_OpMan.pdf
Ours is set to Prog 5. heat=600, Pull=105, Vel=80, Time=250.
Using this we pull needles for injections into mosquito tissues. Hope this is helpful!
The Puller setting is a bit tricky question.I believe that it is not possible to have a standard parameter setting for all the machines.That means your machine setting will be different from my puller setting.The settings are defined by the filament you use.You have to calibrate your own puller setting.For that you need to conduct a Ramp test first-
When a ramp test is executed, the following events take place:
1. The puller increments the HEAT
2. As the HEAT output begins to soften the glass, the puller bars will move apart
3. The heat is then turned off when a certain factory-set Ramp Test velocity is reached
4. The Ramp Test value will be shown on the display
For example ,say a ramp value of 425 volt was obtained using a simple formula provided by Sutter instrument i.e. Heat for pulling =Ramp value +15 a voltage of 440 would be obtained. In the final setting Heat- would be 440 v. The H is the most important setting and you can play around with the rest.Incorrect H would vapourize the filament .I already have burnt quite a few filament in my machine using the wrong setting with out calibrating the machine using the Ramp test.I hope you find the informations useful.
