Hello,

My understanding is that the E value (ideally between 90-110%) gives information about the efficiency of the primers, whereas the R2 value gives information about the linearity of the dilution (so they give different and complementary information). So one could get a really good R2 value but a bad E value. However, I have seen people consider the R2 value as more important than the E value. So for example, if you have a R2 value of 0.998 but a E value of 130%, would you consider that as better that a R2 value of 0.80 with a E value of 99%?
