I am working on bacterial secreted protein, I have cultivated bacteria on non protein minimal medium, then cultures were centrifuged and supernatant filtered though 0.2 micron membrane but I added tricholoroacetic before soudium deoxycholte I am asking if this will affect on my protein extract yield or population?
Sodium Deoxycholate is a detergent and is one of the component of the RIPA buffer to get the proteins embedded in the membrane out in solution. Trochloroacetic acid is a prepitating agent. Since you are analysing just the extracellular media without any cells, sodium deoxycholate should not affect your yield, whether added before TCA or after TCA.

Yes , I just analyze extracellular protein all bacterial cells were separated and and media sterile filtered before precipitation step.
