I am planing an experiment to figure out the effect of an compound for recovery after stroke.
I am wondering whether sham mice group is necessary for this study.
Some reference didn't use sham or others did it.
In my opinion, only two groups (compound-treated mice and compound-non treated mice after stroke) are enough to demonstrate the effect of the compound.
Will reviewer ask the sham result during paper submission process?
A 'sham' group that will be subjected to the stroke-induction procedure (without actually inducing stroke) should be included. This helps to rule out the effect of the procedure itself on the expression of or recovery from stroke.

we are also working on different stroke models like MCAO and BCCAO in rats. It is always necessary to include sham group in your experiments. Sham group results will rule out any changes that will seen due to stress produced during surgical procedures. Sham group will act as a negative control of your experiment.

The sham -operated animal is crucial to rule the harm what the surgery itself can evoke . It would be great to check your compound also in this group of animal to have extra control for effect this compound.
