I am trying to develop leukemia in NOD SCID mice with BCR/ABL over expresssed cell line. But somehow the mice is not showing any signs of the disease. The passage number of cell line is very high. I am wondering whether the passage number of cells has something to do with the failure of experiment. I have injected 5 million cells intravenously after sublethal irrradiation. Any suggestions are welcome.

Good afternoon! Of course, the number of cell line passages affects the result of the experiment. During the passage of a cell line, the cells are senescence, change its morphology, division rate, and survival. It is better to use cells at 2-3 passages.

Yes, the passage number could certainly be a factor in the poor take rate. What are the viability of the cells prior to implantation?

Definitely yes. Passage number matters a lot. U will find an article at ATCC effect of passage number on well studied cell line. Passage number effect the gene expression, tumorigenicity, function etc.
