I want to transduce my target cells with vectors: pLXSN/SV40 LT and pLPCX/hTERT as it was successfully done by Pan et al. (PMID:20620546) or Bian et al (PMID: 15973764) to achieve cells immortality using retroviral packing cells PT67. Further I want to investigate the functionality of particular promoters in the immortalized cells upon stimulation. The problem is I do not have appropriate vector. pLXSN and pLPCX gives resistance for neomycin and puromycin respectively. I would prefer vector giving resistance for other compound (e.g hygromycin or some other). Anyone knows the retroviral vector that has LTRs suitable for PT67 packing cells. I am looking for the vector that does not have constitutive promoter but instead the investigated promoter + lucipherase may be added. It would be great if it had lucipherase in the backbone and only investigated promoter would be needed to be cloned.
Why not. SV40 is well known retrovirus.You have to try the experiment, if you have a properfluminesce reader.
