I am going to do endothelial cell culture from HUVEC. Then i will see several miRNA expression on them after treating endothelial cells with virus infected sera. Right now i dont have my culture media but miRNA primer have arrived. So i was thinking to standardize my primer using PBMC from human blood. I am confused will it be same expression level in PBMC as endothelial cells as my actual work involve endothelial cells or can i use PBMC to standardize primer?

The expression of miRNA is different between cells, but also probably between the same cells under different physiological, pathological and environmental conditions via epigenetic modulations. So, I think that will be unuseful to standardize your primer using PBMC from human blood for their eventual use with endothelial cells.

I do not expect they will be the same since they are different cell phenotypes. But, they may be very similar in some groups of miRNAs.
The expression levels of miRNAs are different even in endothelial cells isolated from different tissues. For example, Umbelical Vein endothelial cells may have different miRNA expression as compared to Lung and brain endothelial cells. Therefore, it is better if you standardize your primer using HUVECs.
