Import of nuclear encoded-mitochondrial proteins into these organelles is coupled to translation. Can you suggest a biochemical method that would allow us to separate ribosomes actively translating mitochondrial proteins?
This could then be followed by polysome profiling and next generation sequencing technology. The objective would be to selectively ask which mitochondrial proteins are being translated actively in cells.
Disclaimer: I am not a translation expert but I think think this would be an interesting experiment...
Some of the imported proteins have UTRs that localize the ribosome on the surface of the mitochondrion for co-translational import. If you isolate mitos, I think these should co-purify (but check that.). This will not get you everything, but might be one neat way to get a hold of some of them.
