I am currently analyzing the antiglycation effects of a few fractions samples from the same plant by conducting BSA-MGO and BSA-Glucose assays. I measured the samples absorbance using 96-well plate at 370 and 420nm. The concentrations of my samples used are 100, 250,500 and 750 ug/ml.
After calculating percent inhibition using formula [(Acontrol - Asample)/(Acontrol)]x100, some fraction samples show decreasing % inhibition with increasing concentrations, some even show negative values of % inhibition at 750 ug/ml.
I searched similar questions and saw a suggested theory of hormetic response (stimulates at low concentration but toxic/inhibits at high concentrations). Is this theory applicable in this situation?
Can anyone suggest other possible reasons or theories regarding the negative percent inhibition values? Or has anyone has faced the same situation before?
