I would like to insert a gene in my cell line. However, I want this inserted gene expressed under particular condition/treatment. IS it possible to create conditionally knock in cells?

briefly, you can use the tTA inducible system:

It's not perfect, has some leakage, and possibly the Tet OFF system is better, so that you will express your gene in the absence of a chemical that might always have secondary effects that add up to your gene expression...
Other possibility, the ProteoTuner expression system:

You can fuse a DD (destabilization domain) to your protein that is therefore constantly produced by very efficiently degraded until you provide a "Shield1 ligand", then in a matter of minutes (about 20 min) your protein will be expressed... Of course in this case you have to keep in mind that you have a fusion protein, some function of your original protein might be affected...
Whichever system, you'll have to live with a drawback...
