sample was first treated with Subcritical water extraction, then liquid products was obtained. After that, for sample preparation before gc-ms analysis, i extract my liquid products with 80% ethanol, took the upper layer, next centrifuge followed by sonicate at 70C. Next, i dry my sample and dissolve it with dioxane, pyridine and N-O,Bis trimethylsilyl acetamide. Last step sonicate and then inject into gcms. Is it my method correct or not?
Depends. What is your target molecule, expected concentration, and sample matrix?
No. Sugar compounds are water soluble (keep away from the non-polar organics). You should also use a 'Thompson Stomacher' to massicate and homogenize the food item.
I would probably use OPA. See the ICUMSA (International Commission of the Uniform Methods of Sugar Analysis) procedure. Something that is more conventional and commonly used.
