I often see in articles that they carry out just an adoptive transfer experiment or just a depletion study, and I was wondering whether you can do both or you only have to do one of them?
I also want to know which one is preferred, or what the pro's and cons are of both of them.

You can perform both depletion and adoptive transfer of T cells in the same system/mouse cohorts! Here is a good example of an paper where depletion follow by T cell transfer was performed (https://iai.asm.org/content/iai/63/10/3827.full.pdf).

).PROS: T cell depletion typically offers an opportunity to define if an effect in vivo is T cell dependent while adoptive transfer of T cells demonstrated the direct potential therapeutic effects of the specific T cells transferred (CAR T cells, antigen specific T cells, pre-stimulated T cells etc). Importantly T cell depletion is a "broader" strategy while adoptive transfer of T cells is a more narrow, directed approach.
CONS: T cell depletion (with anti-CD4 or anti-CD8) can also impact other immune subsets that express CD4 or CD8 but are not "conventional" T cells (NK-T cells, CD4+ Dendritic cells). Adoptive T cell transfer can induce therapeutic effects but you need to have a sufficient number of T cells with the proper activation state and antigen specificity to see significant therapeutic effects
