I've ran a western on mouse ileal lysates and probed for COX-2. 50ug was loaded per sample and the dilution factor was 1:500 for the primary (monoclonal mouse ab) and 1:10000 for the secondary. Upon imaging, I see bands at ~75kDa and around 50kDa. The 75 kDa signal is expected and more prominent than the 50kda signal. However, I did not expect any signal lower than 70kDa, per other papers I've read on COX-2 protein expression. I was told the lower kDa might be a cleaved version of the protein, however, I have not found many papers with a signal close to 50kDa.
