Is there a way to calculate the pIC50 value of a plant extract? As far as I know, in order to calculate pIC50, the IC50 must need to be in molarity, therefore, it's not possible to get the value since we don't know the molecular weight of the total extract.
Well pIC50 is just a statistical term. If you are dealing with extracts, you have a complex mix and therefore you will have a mass concentration of your sample. There is no established rule how to express it like in molar concentration so pIC50 is irrelevant. You can calculate it but it would be unusable for comparisons with other authors that expressed their concentration in a different units.

Technically, you should be able to obtain an IC50 value simply by using relative concentrations like percentage of extract. The problem (or one of them) is that since you are working with an extract, it would be hard to get reproducible results when carrying out the cytotoxic/cytostatic assays to find the IC50. This is mainly due to the differences in extract contents from sample to sample.
Additionally, IC50 is a unit to measure the effect of a inhibitor in a particular biological process. In this case, many molecules could be involved in the inhibition of whatever process you are testing.
