An Anaerobic sludge from digester consists of various anaerobic organism. Is it possible to use media like Nutrient broth, LB broth, MSM,any such basal minimum media to maintain uniform mix culture or to maintain my own culture.
What was the digester fed with originally? The composition of the mixed microbial population will likely change significantly if you maintain it on a media such as nutrient broth or LB. There would still be a mixed community, it just wouldn't be the same as the one originally present in the digester.
I agree with Mark. Why are you not maintaining the cultures independently? whats the purpose of maintaining in mixed cultures.. microbial consortia may be successful or not depending the individual strains behaviours.
If you wish to maintain it in its original and/or initial composition of microbial organisms you should keep it at zero to max. two centigrades in the refrigerator which works for a couple of months. inoculation into any other media will alter the abundance of the consortium as Mark already mentioned.
It could be helpful to know what you intend to do with your anaerobic sludge mixture if you expect from us a more precise reply!!
Fine! If you wish to produce volatile fatty acids such as ???? (which do you have in mind?) you must feed your mixed culture with a carbon- and energy source. A problem may arise: you may loose the bugs you need for this purpose. As a consequence it should be better to isolate first respective producers and use only those in your anaerobic fermentor.
