We are working on histology of some glands using the paraffin method but since the process is too long and we do not have an automatic processing machine, so we do not have any option other than going for an overnight step. If not 70% ethanol than at what point can we leave the sample overnight?

We usually stop in 70% ethanol overnight and we didn't encounter any problems yet. Tho, we usually don't work with samples that are so thin.
Stopping in 100% ethanol might work fine, but it could harden your tissue and make it hard to cut later.
All in all, best would be to give both options a try (if you can) before committing samples that might be impossible to obtain again.
