Hello, I conduct a re-assay on DLD-1 and Vero cell lines.
The result was as I expected, the extract inhibit the proliferation of the DLD-1 but not at Vero.
But as for the morphology of the cell, I couldn't decide if the DLD-1 cell was Apoptosis or Necrosis, since the pics I got were not that good.
Would you please help me decide based on the pics I got?

The pics as you mentioned are not good. Nevertheless, from the pic, I feel it is necrosis because I can see formation of membrane bubbles which is typical of early necrosis. Then there would be disorganization of cytoplasm, homogeneous nuclear condensation and final chromatin disintegration within flattened polygonal cell remnants while still attached to the substratum, which is a characteristic of late necrosis. These characteristics are seen in the pic.
While in apoptosis, you would see severe cell shrinkage, rounding and blebbing of cells followed by chromatin condensation and fragmentation, as well as most important, the cells will detach from the substratum.

I am managing to re-take the pics so I will have better explanation for this extract.
I had my hypothesis that this is a necrosis as well, but yes, still need some expert to help me decide.
I do not know about the microscope, but when I used the microscope with other specimen like fungi and bacteria, the pics were good. but when I do with this cell inside 96 assay plate, the pics just got blurry..
any other advice for re-taking the pics? also, I am considering using other microscope.
I would say its of apoptosis. Cellular blebbing was obvious in your pics. But the quality of your images can be improved. Try staining the cells with Hoechst or other stains to see the shapes of the nuclei.
Besides, keep your cells a bit longer, you would observe more tiny cellular blebs or debris.

Thankyou Ivan Kong , the pics I took were from 18, 24 and 42 hours and I can't take the 48 hours pics due to the cck-8 treatment before the reading with plate reader.
the concentration of the extract were also on 0.2, 0.4 and 0.8 uL and I took the pics as well, but the pics were not that good.
