Is it necessary to remove the N terminal transmembrane domains from Bait protein while contructing vectors?? Why?

I am trying to find interaction among known proteins, I have a bait protein and 8 prey. My bait protein is little large > 1000 aa, which contains 3 transmembrane helix at N terminal. Is it necessary to remove all these aa along with N terminal mitochondrial targeting peptide in Gal4 based Y2H system?? Any reference material for this would be highly appreciated. Thank you.
