I need to make a LiCl/TFE buffer for diluting PPIase substrate (suc-AAPF-pNA) and I was reading the instruction that I have to dry LiCl in vacuum oven for 24 h and dry the TFE with a dry agent such as CaCl before making a fresh buffer. The problem is I don't have a vacuum oven to dry the LiCl in, so I though about skipping the dry step and just use it as it is, but I'm not sure if this might affect the outcome of the experiment "PPIase activity assay"

When the chemicals contain water you will not get the correct molarity by weighing. Depending on your application, this imprecission may not matter. If it does, you could use other means to determine the exact concentration of your stock solutions (e.g., chloride titration with AgNO3 in case of LiCl).
