We used ROC curve analysis to assess the discriminatory power of a microRNA in distinguishing patients from healthy controls. and I want to consul whether is it correct to calculate a ROC curve for data obtained with the -ddCt methods using a calibrator?
Should we calculate a ROC curve using the date obtainde with ddCt instead of -ddCt ?
Not expert in this but I really see no difference in using either ddCt or -ddCt. The ROC curve should give you inverse curve but the way you determine the cut-off will be similar.
I agree to Ru-Jeng, There is no difference in using either -ddct or +ddct (the meaning of the sign is anyway unclear as long as you don't say how the dct and the ddct values were calculated). The only difference concerns the meaning of the ddct values (not the discriminatory power of the ROC analysis!): what does a positive ddct value mean (induction or repression of the gene of interest)?
