It's been a while since I last isolated M1/M2 macrophages but we always used to classifiy M1 as CD11b+CD11c+F4/80+ for M1 and M2 as CD11b+CD11c-F4/80+. Theres a few authors such as CN Lumeng and JM Olefsky who do a lot of M1/M2 research might be an idea to check out some of their papers!

I don't know about surface markers, but IL-12 is for M1 and IL-10 is a phenotype for M2. ELISA kits are available for these.
