I would like to know more about this technique in order to detect protein-protein interaction in 293T cells, is it hard to set up and optimize? What are the drawbacks?

I would say that the the first thing you have to consider is "have you got the good antibodies?". Usually these kits are made to be extremely user friendly, but to find the right high purity, high affinity primary antibodies may require a little time...
Thank you Leonardo for your answer, I think I have the right antibodies, they work well in Immunofluorescence, do you think these kits work well for a broad range of interactions?
I am trying to do a different kind of thing with PLA, so I'm not the best person to tell you. BUT I can put you in contact with a friend who had a positive experience with it and you can discuss the possibilities with him.
