As per the guidelines of different international bodies, total coliform count is the standard parameter for evaluating water microbiological quality. So, culture based quantification is more appropriate.
Obviously DNA from coliforms indicate coliforms have come into the water. For risk consideration one should not forget that it is intact bacteria that might be a risk.
E.g. in Denmark it was in the news that one can measure a lot of DNA of colifrms and pathogenic bacteria in a well maintained public swimming pool while the culture based bacteria detection shows the pool water is microbiologically safe. DNA last a long time in swimming pool water but it is harmless.
