I would like to add BG-azide to cells for SNAP tag pulldown however I don't know whether it is going to be cell permeable. My instinct is to day yes it will be but neither me nor the supplier know whether that is true. I thought I would ask if anyone has tried something similar, even though that is unlikely... I have attached the structures in case that is informative

We in the late Alan C. Sartorelli lab synthesized many O6-Benzylguanine analogs and prodrugs thereof, and found then to be readily taken up by a wide variety of tumor cells.
The molecule is not charged, just somewhat polar, so it should easily pass a lipid bilayer, i.e. a cell membrane. Plus, it might be actively taken up via receptors binding purines. Have you checked the references on the product pages (also from the other SNAPs) in our catalogue?
If in doubt, use the molecule without the PEG spacer, RL-3950. It's more lipophilic. We know from our Biotin-tyramides, that the probe without th PEG spacer permeates, while the one with the PEG doesn't, and thus can be used for selective labeling of intracellular proteins.
