Hello dear colleagues,

I am trying to generate OVA expressing cell lines at the membrane surface in CT26 or B16F10 cells and my first assay are completely unsuccessful (only 4 positive clones for B16 et no clones (150 screened for CT26). The transfection efficiency was good (about 85% and 30% for B16 and CT26 respectively) We use a DNA encoding truncated OVA fused to Transferrin receptor membrane anchoring domain and it works in EL4 or when transiently expressed in HEK or B16F10. Is anybody has had a similar issue for obtaining membrane-bound protein in B16 or CT26?
