High salt is important as I recall. The abundance of cytoskeletal and membrane/membrane associated proteins is an issue with skeletal muscle cells.

A high salt buffer will help precipitate actin so you can pellet it during clarification of your lysates (usually 15+ min at 15,000xg) before SDS-PAGE and immunoblotting.
